PD-L1 upregulation is associated with activation of the DNA double-strand break repair pathway in patients with colitic cancer

چکیده
Abstract Ulcerative colitis (UC) is a DNA damage-associated chronic inflammatory disease; the double-strand break (DSB) repair pathway participates in UC-associated dysplasia/colitic cancer carcinogenesis. The DSB/interferon regulatory factor-1 (IRF-1) can induce PD-L1 expression transcriptionally. However, association of PD-L1/DSB/IRF-1 with sporadic colorectal (SCRC), and cancer, remains elusive. Therefore, we investigated significance PD-L1/DSB using samples from 17 SCRC 12 UC patients rare cases by immunohistochemical analysis. We compared between determined CD8+ T-cell/DSB/IRF-1 axis cancer. was higher than normal mucosa or SCRC, CD8-positive T lymphocytes SCRC. Moreover, upregulation associated γH2AX (DSB marker) IRF-1 but not Multicolour immunofluorescence staining validated γH2AX/IRF-1/PD-L1 co-expression colitic tissue sections. Thus, immune cell-induced inflammation might activate DSB/IRF-1 axis, potentially serving as primary mechanism

متن کاملDNA double-strand break repair
The integrity of genomic DNA is crucial for its function. And yet, DNA in living cells is inherently unstable. It is subject to mechanical stress and to many types of chemical modification that may lead to breaks in one or both strands of the double helix. Within the cell, reactive oxygen species generated by normal respiratory metabolism can cause double-strand breaks, as can stalled DNA repli...
متن کاملMismatch-repair protein MSH6 is associated with Ku70 and regulates DNA double-strand break repair
MSH6, a key component of the MSH2-MSH6 complex, plays a fundamental role in the repair of mismatched DNA bases. Herein, we report that MSH6 is a novel Ku70-interacting protein identified by yeast two-hybrid screening. Ku70 and Ku86 are two key regulatory subunits of the DNA-dependent protein kinase, which plays an essential role in repair of DNA double-strand breaks (DSBs) through the non-homol...
متن کاملDNA double-strand break repair pathway choice in Dictyostelium.
DNA double-strand breaks (DSBs) can be repaired by homologous recombination (HR) or non-homologous end joining (NHEJ). The mechanisms that govern whether a DSB is repaired by NHEJ or HR remain unclear. Here, we characterise DSB repair in the amoeba Dictyostelium. HR is the principal pathway responsible for resistance to DSBs during vegetative cell growth, a stage of the life cycle when cells ar...
متن کاملNampt is involved in DNA double-strand break repair
DNA double-strand break (DSB) is the most severe form of DNA damage, which is repaired mainly through high-fidelity homologous recombination (HR) or error-prone non-homologous end joining (NHEJ). Defects in the DNA damage response lead to genomic instability and ultimately predispose organs to cancer. Nicotinamide phosphoribosyltransferase (Nampt), which is involved in nicotinamide adenine dinu...
